Difference between FlowBox and SimpleBox ?

The main difference is really the automatic video playback workflow that is built into FlowBox. SimpleBox works differently by you applying for example the Vimeo embed code in a SimpleBox container.
Flowbox will read your page and determine if it is Vimeo, YouTube or H.264 on the fly.
